The structural features incorporated in the main (1-6)-linked backbone chain, the acid-stable side chains attached to the backbone chain and the acid-labile phosphate-linked side chains are characterized by 1D proton NMR and 2D COSY NMR. The side chains can be composed of α- and ß-linked (1-2)- and (1-3)-glycosidic linkages. Based upon NMR analysis, it is straight-forward to characterize mannan structural features and determine changes in the amount of these structural features by comparison with a reference mannan spectrum or one of the mannan spectra in your study. We have characterized structures of mannans isolated from Saccharomyces cerevisiae (1), Candida albicans (1,2), C. glabrata (3) and C. auris (4).
